摘要:
目的:探讨AURKA蛋白激酶在三阴乳腺癌干细胞形成血管拟态中的作用。方法:用无血清悬浮培养法从三阴乳腺癌细胞MDA-MB-231分离获得干细胞,RT-PCR检测干细胞转录因子c-myc、sox-2的表达状态;三维培养观察干细胞形成血管能力;Western blot 检测AURKA蛋白激酶在干细胞和常规培养细胞的表达;利用干细胞进行三维培养并加入AURKA蛋白激酶抑制剂观察对成血管能力的影响。结果:(1)利用干细胞无血清悬浮培养可获得MDA-MB-231干细胞,其干细胞转录因子c-myc、sox-2表达高于常规培养细胞。(2)AURKA蛋白激酶在乳腺癌干细胞球中较常规培养细胞表达升高。(3)干细胞三维培养可形成血管管道,加入AURKA蛋白激酶抑制剂后成血管能力减弱。结论:AURKA蛋白激酶可促进三阴乳腺癌干细胞形成血管拟态,抑制其表达能减弱形成血管拟态能力。
Abstract:
Objective: To investigate the role of AURKA protein kinase in the formation of vascular mimicry in triple-negative breast cancer stem cells. Methods: The serum-free suspension culture of breast spheres from triple-negative breast cancer cells MDA-MB-231 cells were isolated, the expression of stem cell transcription factor status of c-myc and sox-2 were detected by RT-PCR. The formation of vascular stem cells were observed by three-dimensional culture. The expression of AURKA protein kinase from stem and normal cells were detected by western blot. Three-dimensional culture and AURKA protein kinase inhibitor were used to detect the formation ability of blood vessels. Results: (1) The expression of stem cell transcription factor status of c-myc and sox-2 were highter than conventional cell culture in MDA-MB-231 stem cells obtained from the serum-free suspension culture. (2) The expression of AURKA protein kinase were higher than conventional cell culture in MDA-MB-231 stem cells obtained from the serum-free suspension culture.(3) Stem cells contributed to the formation of vascular pipeline, AURKA protein kinase inhibitor diminished the formation of vascular. Conclusion: AURKA plays a key role in the formation of vascular mimicry in triple-negative breast cancer stem cells,inhibiting its expression and weakening the ability of the formation of vascular mimicry.
