摘要:
目的:观察补充维生素D对严重烫伤小鼠血清炎性因子浓度变化的影响。方法:(1)随机将88只小鼠分为实验组和对照组各40只,空白对照组8只。(2)实验组小鼠烫伤后即刻给予1,25(OH)2D3 4 μg/kg+0.6 mL花生油灌胃,每日1次,至处死前;对照组烫伤小鼠0.6 mL花生油灌胃,每日1次,至处死前。(3)于伤后6、12、24、48、96 h各组分别随机选择8只小鼠,用ELISA法检测血清TNF-α、IL-1β、IL-4、IL-10含量。(4)于伤后96 h取两组小鼠肠系膜、肝和脾淋巴结组织分别制成匀浆,做48 h细菌培养计算菌落形成单位数量。结果:(1)烫伤后12、24、48、96 h,实验组和对照组小鼠血清TNF-α、IL-1β、IL-4、IL-10浓度分别高于空白对照组水平(P<0.05)。(2)烫伤后24、48、96 h,实验组小鼠血清TNF-α和IL-1β浓度分别低于对照组水平(P<0.05);烫伤后12、24、48、96 h,实验组小鼠血清IL-4和IL-10浓度分别高于对照组水平(P<0.05)。(3)伤后96 h实验组小鼠肠系膜、肝和脾淋巴结组织菌落形成单位量分别低于对照组(P<0.05)。结论:严重烫伤小鼠伤后补充维生素D能够降低TNF-α和IL-1β浓度、提高IL-4和IL-10浓度,并减少肠系膜、肝和脾淋巴结组织菌落形成单位数量
Abstract:

Objective To explore the effect of vitamin D supplementation on the changes of serum inflammatory cytokines in severely scalded mice. Methods(1) Eighty-eight mice were assigned into 3 groups randomly: treatment group(40), control group(40) and blank group(8). (2)Mice in treatment group were immediately fed 0.6 mL peanut oil with1,25(OH)2D3(4 μg/kg) once a day post-burn, and mice in control group received only 0.6mL peanut oil without 1,25(OH)2D3. (3)The levels of serum TNF-α, IL-1β, IL-4 and IL-10 were determined respectively by ELISA at the time of 6, 12, 24, 48 h and 96h post-burn. (4)The colony-forming units of liver, spleen and mesenteric lymph nodes of mice in two groups were calculated respectively at 96 h post-burn. Results:(1)The serum TNF-α, IL-1β, IL-4 and IL-10 levels in treatment and control group were significantly higher than those in blank group at the time of 12, 24, 48 h and 96h post-burn(P<0.05). (2)The serum TNF-α and IL-1β levels in treatment group were lower than those in control group at the time of 24, 48 and 96 h post-burn(P<0.05), and the serum IL-4 and IL-10 levels in mice in treatment group were higher than those in control group at the time of 12, 24, 48 h and 96 h post-burn(P<0.05). (3)The numbers of colony-forming units of liver, spleen and mesenteric lymph nodes of mice in treatment group were lower than those in control group at 96 h post-burn(P<0.05). ConclusionSupplementing vitamin D could reduce the levels of serum TNF-α and IL-1β, and increase the level of serum IL-4 and IL-10 in severely scalded mice. And it could reduce the numbers of colony-forming units of liver, spleen and mesenteric lymph nodes of mice post-burn.
