Effect of vitamin D supplementation on the changes of serum inflammatory cytokines in severely scalded mice

Objective :To explore the effect of vitamin D supplementation on the changes of serum inflammatory cytokines in severely scalded mice. Methods:(1) Eighty-eight mice were assigned into 3 groups randomly: treatment group(40), control group(40) and blank group(8). (2)Mice in treatment group were immediately fed 0.6 mL peanut oil with1,25(OH)2D3(4 μg/kg) once a day post-burn, and mice in control group received only 0.6mL peanut oil without 1,25(OH)2D3. (3)The levels of serum TNF-α, IL-1β, IL-4 and IL-10 were determined respectively by ELISA at the time of 6, 12, 24, 48 h and 96h post-burn. (4)The colony-forming units of liver, spleen and mesenteric lymph nodes of mice in two groups were calculated respectively at 96 h post-burn. Results:(1)The serum TNF-α, IL-1β, IL-4 and IL-10 levels in treatment and control group were significantly higher than those in blank group at the time of 12, 24, 48 h and 96h post-burn(P<0.05). (2)The serum TNF-α and IL-1β levels in treatment group were lower than those in control group at the time of 24, 48 and 96 h post-burn(P<0.05), and the serum IL-4 and IL-10 levels in mice in treatment group were higher than those in control group at the time of 12, 24, 48 h and 96 h post-burn(P<0.05). (3)The numbers of colony-forming units of liver, spleen and mesenteric lymph nodes of mice in treatment group were lower than those in control group at 96 h post-burn(P<0.05). Conclusion:Supplementing vitamin D could reduce the levels of serum TNF-α and IL-1β, and increase the level of serum IL-4 and IL-10 in severely scalded mice. And it could reduce the numbers of colony-forming units of liver, spleen and mesenteric lymph nodes of mice post-burn.
