摘要:
目的:探讨氢气对酵母多糖(ZY)诱导的脓毒症小鼠存活率和肝脏损伤的影响及相关机制。方法:雄性ICR野生型小鼠120只,体质量20~25 g,5周龄,采用随机数字表法分为4组(n=30):正常对照组(A组)、氢气组(B组)、ZY组(C组)、ZY+氢气组(D组)。C和D组小鼠腹腔注射ZY制备脓毒症模型;B和D组小鼠在腹腔注射后的1 h和6 h时分别吸入2%氢气1 h。每组取20只观察7 d内小鼠生存情况。另40只动物,实验后24 h下腔静脉采血,取肝脏组织,检测血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)水平及进行肝脏组织病理评分,测定血清和肝脏超氧化物歧化酶(SOD)及8-异前列腺素F2α(8-iso-PGF2α)水平。结果:与A组相比,C组动物存活率降低,血清AST、ALT水平及肝脏病理评分升高,血清和肝脏组织SOD活性降低,而8-iso-PGF2α水平明显升高(P<0.05);与C组相比,D组存活率提高,血清AST和ALT水平及肝脏病理评分降低,血清和肝脏组织中SOD活性升高,而 8-iso-PGF2α水平降低(P<0.05)。结论:氢气可改善脓毒症小鼠存活率,减轻肝脏损伤,其机制可能与抑制氧化应激反应有关。
Abstract:
Objective: To investigate the protective effects of hydrogen gas (H2) on survival rate and liver injury in zymosan (ZY)-induced septic mice. Methods: One hundred and twenty male ICR mice, aged 5 weeks, weighing 20-25 g, were randomly divided into 4 groups (n =30 each): control group (group A), H2 group (group B), ZY group (group C) and ZY + H2 group (group D). Septic mice were produced by peritoneal injection of ZY in groups C and D. Mice in groups B and D received 1 h inhalation of 2% H2 at 1 and 6 h after normal saline or ZY injection. Twenty animals in each group were selected and observed for the 7 d survival rates. The left 10 animals in each group were sacrificed at 24 h after injection. Venous blood samples and liver tissues were obtained to determine the levels of superoxide dismutase (SOD) and 8-iso-prostaglandin F2alpha (8-iso-PGF2α). Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were tested, meanwhile liver histopathology were examined. Results: Compared with group A, the animals in group C showed a significant decrease of 7 d survival rate, liver injury (the increase of serum ALT, AST and histopathological scores), the decrease of SOD activities and increase of 8-iso-PGF2α levels in serum and liver tissues (P<0.05). Compared with group C, the animals in group D showed a significant increase of 7 d survival rate, improvement of liver injury (the increase of serum ALT, AST and histopathological scores), increase of SOD activities and decrease of 8-iso-PGF2α levels in serum and liver tissues (P<0.05). Conclusion: Hydrogen can improve the survival rate and liver injury in septic mice via inhibiting oxidative stress.
