摘要:
目的:探讨宣肺通腑塌渍方外治对肺炎支原体(MP)肺炎小鼠肠道菌群的影响。方法:20只SPF级C57BL/6野生型小鼠随机分为4组,即对照组、模型组、宣肺通腑外治组以及阿奇霉素组,每组5只。制备重组社区获得性呼吸窘迫综合征(CARDS)毒素,使用CARDS毒素制备MP肺炎模型,苏木精-伊红染色法(HE)观察小鼠肺组织病理变化,16SrRNA(核糖体RNA)菌群测序分析肠内容物中肠道菌群变化。结果:模型组肺组织病理切片可见炎性浸润增加,经宣肺通腑外治干预后炎性浸润得到改善;与模型组[(13.6±1.1)分]相比,宣肺通腑外治组病理评分[(11.0±1.4)分]降低,差异具有统计学意义(P<0.05)。Alpha多样性分析,与模型组相比,宣肺通腑外治组Chao指数降低(H=1.423,P=0.49),而Simpson指数(H=3.577,P=0.17)和Shannon指数(H=2.923,P=0.23)均升高,但3组比较均无统计学差异(P>0.05)。Beta多样性分析,宣肺通腑外治组与对照组样本点部分重叠,较为接近,表明宣肺通腑外治后可调整肠道菌群。Adonis分析证实各组间菌落结构存在显著差异(R2=0.216,P=0.001)。肠道菌群组成结构分析发现,拟杆菌门为优势菌门。与对照组相比,模型组拟杆菌门相对丰度下调;与模型组相比,宣肺通腑外治组相对丰富度上调(F=6.287,P<0.05)。鼠杆菌属在3组间有统计学差异(F=4.64,P<0.05)。结论:宣肺通腑塌渍方外治减轻MP肺炎小鼠肺损伤,可能与恢复肠道菌群稳态有关。
Abstract:
Objective: To investigate the effect of external treatment with the Xuanfei Tongfu Soaking Formula on gut microbiota in Mycoplasma pneumoniae(MP)-infected mice. Methods: Twenty SPF-grade C57BL/6 wild-type mice were randomly divided into four groups: control group, model group, Xuanfei Tongfu external treatment group and azithromycin group, with 5 mice in each group. Recombinant community-acquired respiratory distress syndrome(CARDS) toxin was prepared and used to establish the MP pneumo-nia model. Pathological changes in mouse lung tissue were observed by hematoxylin-eosin(HE) staining, and alterations in the gut microbiota of intestinal contents were analyzed by 16S rRNA(ribosomal RNA) sequencing. Results: Histopathological sections of lung tissue from the model group showed increased inflammatory infiltration, which was ameliorated after intervention with Xuanfei Tongfu external therapy. The pathological score in the Xuanfei Tongfu external treatment group(11.0±1.4) was significantly lower than that in the model group(13.6±1.1), with a statistically significant difference between the two groups(P<0.05). Alpha diversity analysis showed that compared with the model group, the Xuanfei Tongfu external treatment group had a decreased Chao index(H=1.423, P=0.49), while the Simpson index(H=3.577, P=0.17) and Shannon index(H=2.923, P=0.23) both increased. However, there was no sta-tistical difference among the three groups(all P>0.05). Beta diversity analysis showed that the sample points of the Xuanfei Tongfu external treatment group and the control group partially overlapped and were relatively close, indicating that the gut microbiota could be adjusted after Xuanfei Tongfu external treatment, and Adonis analysis confirmed significant differences in colony structure among groups(R2=0.216, P=0.001). Statistical analysis of the relative abundance of gut microbiota at the phylum level revealed that Bac-teroidota were the dominant phyla. Compared with the control group, the model group showed a down-regulation in the relative abun-dance of Bacteroidota. In contrast, the Xuanfei Tongfu external treatment group exhibited an up-regulation in the relative abundance of Bacteroidota compared to the model group(F=6.287, P<0.05),there was a statistically significant difference related to Muribacu-lum among the three groups(F=4.64, P<0.05). Conclusion: The external application of the Xuanfei Tongfu Soaking Formula allevi-ates lung injury in mice with MP pneumonia, which may be associated with the restoration of gut microbiota homeostasis.
