摘要:
目的:通过积雪草酸与羟丙基-β-环糊精(HP-β-CD)的络合作用来提高积雪草酸在水中的溶解度。方法: 用红外光谱(FTIR)、X-射线衍射(XRD)、差示扫描热仪(DSC)和扫描电镜(SEM)对积雪草酸/ HP-β-CD络合物的性质进行测试。结果:研究发现当积雪草酸: HP-β-CD为1:2时,积雪草酸在水中的溶解度达到2 100 μg/mL,是积雪草酸在水中溶解度的17倍。 红外光谱图和X-射线衍射图谱证实了积雪草酸/ HP-β-CD络合物以固体状态存在。扫描电镜、差示扫描热和X-射线衍射图谱表明积雪草酸以无定形形态存在于络合物中。结论:羟丙基-β-环糊精通过与积雪草酸络合,能有效增加积雪草酸的溶解度。
Abstract:
Objective: To improve the solubility of asiatic acid, a poorly water soluble drug, by combining with hydroxypropyl-β-cyclodextrin (HP-β-CD). Methods: The asiatic acid/HP-β-CD solid complex system was characterized by Fourier Transform Infrared Spectroscopy (FTIR), X-ray diffraction (XRD), Differential Scanning Calorimetry (DSC)and Scanning Electron Microscopy(SEM). Results: The effect of asiatic acid: HP-β-CD feed ratio by molar on the aqueous solubility was investigated and the aqueous solubility of asiatic acid reached 2 100 μg/mL when the molar ratio of asiatic acid to HP-β-CD was 1:2. The aqueous solubility of asiatic acid was increased by 21-fold in asiatic acid/HP-β-CD solid complex. The FTIR and XRD spectra of asiatic acid/HP-β-CD solid complexes showed that Asiatic acid could form inclusion complex with HP-β-CD in solid status. The SEM, DSC and XRD spectra of asiatic acid/HP-β-CD solid complexes indicated asiatic acid was in amorphous status. Conclusion: Hydroxypropyl - β - cyclodextrin  can effectively increase the solubility of asiatic acid by combining with asiatic acid.
