摘要:
目的:探讨舒芬太尼对宫颈癌HeLa细胞作用机制。方法:体外培养HeLa细胞,分为舒芬太尼低、中、高剂量组(10、20、40 μg/mL舒芬太尼)、激活剂组(40 μg/mL舒芬太尼+100 ng/mL RhoA激活剂PMA)、抑制剂组(40 μg/mL舒芬太尼+5 μg/mL RhoA抑制剂C3转移酶)和对照组;CCK-8法测定HeLa细胞增殖;划痕试验测定HeLa细胞迁移;Transwell测定HeLa细胞侵袭;流式细胞术检测HeLa细胞凋亡;Western Blotting检测RhoA、ROCK蛋白及E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)表达。结果:与对照组比较,舒芬太尼低、中、高剂量组细胞存活率、细胞侵袭数目、划痕愈合率及N-cadherin、Vimentin、RhoA、ROCK表达下降,凋亡率和E-cadherin蛋白表达增加(q=4.695~30.652,均P<0.05);与舒芬太尼高剂量组比较,激活剂组细胞存活率、细胞侵袭数目、划痕愈合率及N-cadherin、Vimentin、RhoA、ROCK表达上升,凋亡率和E-cadherin表达下降(q=4.378~14.454,均P<0.05),抑制剂组细胞存活率、细胞侵袭数目、划痕愈合率及N-cadherin、Vimentin、RhoA、ROCK蛋白表达下降,凋亡率和E-cadherin蛋白表达增加(q=4.501~7.545,均P<0.05)。结论:舒芬太尼通过下调RhoA/ROCK信号通路,阻碍宫颈癌HeLa细胞的EMT和增殖,诱导细胞凋亡。
Abstract:
Objective:To explore the mechanism of the effect of sufentanil on cervical cancer HeLa cells. Methods: HeLa cells were cultured in vitro and grouped into low, medium, high dose group of sufentanil (10, 20, 40 μg/mL sufentanil), activator group (40 μg/mL sufentanil+100 ng/mL RhoA activator PMA), inhibitor group (40 μg/mL sufentanil+5 μg/mL RhoA inhibitor C3 transferase) and control group; CCK-8 assay was applied to detect the proliferation of HeLa cell; scratch test was applied to detect the migration of HeLa cell. Transwell was used to detect the invasion of HeLa cell. Flow cytometry was applied to detect the apoptosis of HeLa cell. The expression of RhoA, ROCK, E-cadherin, N-cadherin, and Vimentin protein were detected by Western Blotting. Results: Compared with the control group, the cell survival rate, number of cell invasions, scratch healing rate, and expression of N-cadherin, Vimentin, RhoA, and ROCK proteins were reduced in the low, medium, and high dose groups of sufentanil, the apoptosis rate and expression of E-cadherin protein were increased (q=4.695-30.652, all P<0.05). Compared with the sufentanil high dose group, the cell survival rate, number of cell invasions, scratch healing rate, and expression of N-cadherin, Vimentin, RhoA, and ROCK proteins in the activator group were obviously increased, the apoptosis rate and expression of E-cadherin protein were reduced (q=4.378-14.454, all P<0.05). The cell survival rate, number of cell invasions, scratch healing rate, and expression of N-cadherin, Vimentin, RhoA, and ROCK proteins in the inhibitor group were reduced, the apoptosis rate and expression of E-cadherin protein were increased (q=4.501-7.545, all P<0.05). Conclusion: Sufentanil inhibits EMT and the proliferation of cervical cancer HeLa cells and induces cell apoptosis by downregulating the RhoA/ROCK signaling pathway.
