摘要:

目的: 观察特异性免疫治疗(SIT)对特应性皮炎(AD)模型小鼠细胞因子的调控,探讨其在免疫方面的作用机制。方法 用尘螨溶液诱导BALB/C小鼠形成过敏体质,再分别用0.5%丙酮和卵清蛋白诱导产生AD模型。用阿罗格脱敏试剂行SIT 1周。分别用Elisa法及Real Time RT-PCR法观察SIT前后小鼠血清IL-4、IL-10、IFN-γ水平及脾脏组织中IL-4 mRNA、IL-10mRNA、IFN-γmRNA表达的变化。结果: 治疗前较正常对照组,血清IL-4、IL-10水平升高,IFN-γ水平降低;脾脏组织中IL-4 mRNA、IL-10mRNA水平升高,IFN-γmRNA水平降低(P均<0.01);SIT后较治疗前血清IL-4、IL-10水平降低,IFN-γ水平升高;脾脏组织中IL-4 mRNA、IL-10mRNA水平下调,IFN-γmRNA水平上调(P均<0.01)。结论:SIT可纠正AD小鼠Th1/Th2细胞因子平衡失调,对除尘螨外丙酮及卵清蛋白引起的特应性皮炎小鼠均有良好的治疗作用。

Abstract:
Objective: To study the effect of specific immunotherapy on cytokines in atopic dermatitis (AD) mice model. Methods: The atopic dermatitis mice was established by sensitization with dust mite allergy solution, and then were treated with 0.5% DNCB or ovalbumin on the skin. The control group was treated with PBS. The remaining mice were treated with specific immunotherapy for one week. The serum levels of IL-4, IL-10 and IFN-γwere detected by Enzy-Linked immunosorbent assay (Elisa). The mRNA levels of IL-4, IL-10 and IFN-γin spleen tissues of AD mice were detected by recerse-transcrition polymerase chain reaction (RT-PCR). Results: Compared with the normal controls, serum levels of IL-4 and IL-10 were increased, and IFN-γwas reduced in AD mice. The IL-4 mRNA and IL-10 mRNA levels of AD mice were up-regulated, the IFN-γmRNA level was down-regulated. After SIT treatment, the expressions of IL-4 and IL-10 were reduced, and the expression of IFN-γwas increased. Conclusion: SIT could correct the imbalance of Th1/Th2 cytokines in mice with AD and have therapeutic effect on allergic dermatitis mice induced by different allergens.
