摘要:
目的: 探讨人工发酵虫草菌粉对糖尿病大鼠肾脏白细胞介素17(IL-17)、p38 MAPK和NF-κB表达的影响。方法:雄性SD大鼠45只,随机选择15只为正常对照组(NC组),其余30只大鼠通过一次性尾静脉注射链脲佐菌素(STZ,45 mg/kg)诱导建立糖尿病模型,造模成功的28只大鼠随机分为糖尿病组(DM组)和人工发酵虫草菌粉治疗组(虫草组,1 g/(kg·d),每组各14只。虫草组给予每日1次人工发酵虫草菌粉溶液灌胃,NC组和DM组给予等体积生理盐水灌胃。18周末取材。计算肾重指数,检测血尿素氮(BUN)、肌酐(Scr)、24 h尿微量白尿蛋白(24 h UMA)。HE染色观察肾脏组织形态学变化;Masson染色观察肾脏纤维化程度;电镜观察各组大鼠肾小球超微结构改变;免疫组化法比较各组大鼠肾脏IL-17、p38 MAPK、NF-κB的表达水平。结果:(1) DM组肾重指数、BUN、Scr、24 h UMA较NC组明显升高,虫草组肾重指数、BUN、Scr、24 h UMA较DM组明显降低。(2) DM组肾脏纤维化明显,虫草组大鼠肾脏纤维化较DM组明显减轻。(3)电镜下,DM组大鼠肾小球毛细血管基底膜增厚,部分足细胞突起融合,系膜细胞增生;虫草组大鼠肾小球病变较DM组明显减轻。(4) DM组肾脏IL-17、p38 MAPK、NF-κB表达均显著上升,人工发酵虫草菌粉干预降低了肾脏IL-17、p38 MAPK、NF-κB的表达。结论:肾脏IL-17、p38 MAPK和NF-κB表达增多在糖尿病肾病发病过程中起到重要作用。人工发酵虫草菌粉可能通过减少肾脏IL-17、p38 MAPK、NF-κB的表达来改善糖尿病肾病。
Abstract:
Objective: To investigate the effect of artificial fermentation cordyceps -powder on the expression of interleukin (IL)-17, p38 MAPK, NF-κB in kidneys of diabetic rats. Methods: A total of 45 male SD rats were randomly divided into normal control group (NC, n =15) and experimental group (n =30). Diabetes was induced by tail vein injection with streptozotocin (STZ, 45 mg/kg). The well-established 28 diabetic model rats were then randomly divided into diabetes group (DM, n =14) and artificial fermentation cordyceps powder administration group (CM, n =14). The rats in group CM were given administration of artificial fermentation cordyceps powder by gavage with a single dose of 1 g/(kg·d )for 18 weeks, while NC group and DM group were given 0.9% NaCl in the same way. All rats were sacrificed after 18 weeks. The ratios of kidney/body weight were calculated. Blood urea nitrogen (BUN), creatinine (Scr) and 24-hour urine albumin(24 h UMA) were detected. The pathological changes in kidneys were studied by HE staining under electron microscope. The degree of renal fibrosis was evaluated by Masson staining. The expression of IL-17, p38 MAPK, NF-κB in kidneys was detected using immunohistochemistry. Results: (1)The blood BUN, Scr and 24 h UMA were significantly higher in group DM than in group NC, and those in group CM were lower compared with group DM.(2) Renal collagen fibers proliferation significantly increased in group DM compared with in group NC, and those in group CM significantly decreased compared with group DM. (3) In group DM, Irregular thicking of glomerular basement membrane, fusion of epithelial cells foot processes and mesangial expansion were observed by electron microscope. The above-mentioned pathological changes were improved in group CM. (4) The expressions of IL-17, p-p38 MAPK, NF-κB in the kidneys significantly increased in group DM than in group CM, and those in group CM were significantly lower than group DM. Conclusion: The increasing expression of IL-17, p38 MAPK, NF-κB in kidneys play a potential role in diabetic nephropathy. Artificial fermentation cordyceps powder can improve diabetic nephropathy through inhibiting the expression of IL-17, p38 MAPK, NF-κB in kidneys.
