摘要:
目的:探究组蛋白去乙酰化酶6(HDAC6)调控中性粒细胞减轻沙眼衣原体呼吸道感染肺组织的炎症机制。方法:野生型C57BL/6(WT)小鼠和HDAC6缺陷(HDAC6-/-)小鼠以鼻腔吸入沙眼衣原体小鼠肺炎菌株(C.muridarum)建立小鼠沙眼衣原体呼吸道感染模型,于感染后0、7 d采集各组小鼠肺组织。采用吉姆萨染色检测中性粒细胞百分比,免疫荧光染色观察肺组织中性粒细胞的分布与表达,流式细胞术检测中性粒细胞百分比、绝对数和活化水平,实时荧光定量PCR(qPCR)检测肺组织中性粒细胞趋化性细胞因子及相关促炎因子mRNA表达水平。结果: C.muridarum呼吸道感染后,与WT小鼠相比,HDAC6-/-小鼠中性粒细胞向肺组织的募集以及活性显著增强(t=6.956、7.705、4.032、12.93、7.829、4.448、3.307,均P<0.05),肺组织角质细胞趋化因子(KC)、巨噬细胞炎症蛋白2(MIP-2)和CXC 趋化因子受体 2(CXCR2)及基质金属蛋白酶8(MMP8)、S100钙结合蛋白A8(S100A8)、肿瘤坏死因子(TNF)-α和白细胞介素(IL)-1β的mRNA表达均上调(t=6.584、3.953、6.437、3.886、5.129、3.317、3.164,均P<0.05)。结论: C.muridarum呼吸道感染中,HDAC6通过降低中性粒细胞募集和活化,减轻感染后肺组织炎症反应,发挥免疫保护作用。
Abstract:
Objective: To investigate the mechanism by which histone deacetylase 6 (HDAC6) regulates neutrophils to attenuate pulmonary inflammation in Chlamydia muridarum respiratory infection. Methods: Wild-type (WT) C57BL/6 mice and HDAC6-deficient (HDAC6-/-) mice were intranasally inoculated with C. muridarum to establish a respiratory infection model. Lung tissues were collected from each group at 0 and 7 days post-infection. Giemsa staining was performed to determine the percentage of neutrophils. Immunofluorescence (IF) staining was used to detect the distribution and expression of neutrophils in lung tissues. Flow cytometry was used to determine the percentage, absolute count, and activation level of neutrophils. Real-time quantitative PCR (qPCR) was performed to measure the mRNA expression levels of neutrophil chemotactic cytokines and related pro-inflammatory factors in lung tissues. Results: After C. muridarum respiratory infection, compared to WT mice, HDAC6-/- mice exhibited significantly enhanced recruitment neutrophils and activation of neutrophils in lung tissues (t=6.956, 7.705, 4.032, 12.93, 7.829, 4.448, 3.307, all P<0.05). The mRNA expression levels of keratinocyte chemokine (KC), macrophage inflamma-tory protein 2 (MIP-2), CXC chemokine receptor 2 (CXCR2) and matrix metalloproteinase (MMP)8, S100 calcium-binding protein A8(S100A8), tumor necrosis factor α (TNF-α) and interleukin-1 β (IL-1 β) in lung tissues were all upregulated (t=6.584, 3.953, 6.437, 3.886, 5.129, 3.317, 3.164, all P<0.05). Conclusion: HDAC6 alleviates pulmonary in-flammation in C. muridarum respiratory infection by reducing neutrophil recruitment and activation , thereby playing an immune-protective role.
