摘要:
目的:建立HPLC同时测定复方甘草胶囊中间体浸膏中苦参碱和氧化苦参碱的含量,为制备胶囊提供依据。方法:色谱柱为Kromasil-NH2柱(250 mm×4.6 mm, 5 μm),流动相为乙腈-无水乙醇-3%磷酸(80:10:10 V/V/V),流速1.0 mL/min,检测波长220 nm,柱温30 ℃。结果:苦参碱在5.12 ~ 30.72 μg/mL(r=0.999 5),氧化苦参碱在5.16 ~ 30.96 μg/mL(r=0.999 5)浓度范围内与峰面积线性关系良好,苦参碱平均回收率99.63%,RSD为0.62%(n=6),氧化苦参碱平均回收率99.81%,RSD为0.72%(n=6)。结论:该方法准确,稳定性好,重复性高,可用于复方甘草胶囊中间体浸膏的质量控制,为胶囊制备提供依据。
Abstract:
Objective: To establish an HPLC method for the determination of matrine and oxymatrine in extraction of compound Liquorice capsules,and to provide the foundation to prepare capsules. Methods: The HPLC was carried out on Kromasil-NH2 column(250 mm×4.6 mm, 5 μm), and mobil phase was acetonitrile-absolute alcohol-3%phosphoric acid(80:10:10,V/V/V).The flow rate was 1.0 mL/min.Detection wavelength was 220 nm.The temperature of column was 30 ℃. Results: The linear rangers of matrine was 5.12 - 30.72 μg/mL(r=0.999 5) ,and oxymatrine was 5.16 - 30.96 μg/mL(r=0.999 5).The average recovery rate of matrine was 99.63% with RSD 0.62%(n=6),and oxymatrine was 99.81% with RSD 0.72% (n=6).ConclusionThis method is accurate, stable and repeatable. It can be used to control the quality of extraction of compound Liquorice capsules ,and to provide the basis to prepare capsules.
