?目的: 探究阿霉素肾病大鼠血清白细胞介素(IL)-17、IL-6的动态变化意义及泼尼松通过2种因子对阿霉素肾病的干预机制。方法: 将81只大鼠随机分为空白对照组、模型组、泼尼松干预组,每组27只,通过分次尾静脉注射法建立阿霉素肾病大鼠模型,检测4、6、8周末大鼠血生化、24 h尿蛋白定量及尿肌酐变化,计算内生肌酐清除率;应用 ELISA检测大鼠血清IL-17、IL-6水平的变化。结果: 实验第4周末,与空白对照组比较,模型组和泼尼松干预组大鼠出现大量蛋白尿、低白蛋白血症和高脂血症(P<0.05),提示模型建立成功;模型组和泼尼松干预组大鼠血清IL-17、IL-6水平高于空白对照组。实验第6、8周末,与模型组比较,泼尼松干预组24 h尿蛋白定量、血肌酐、总胆固醇显著降低 (P<0.05),血清总蛋白、白蛋白、内生肌酐清除率显著升高(P<0.05);泼尼松干预组大鼠血清IL-17、IL-6水平明显低于模型组(P<0.05)。结论: IL-17、IL-6参与了阿霉素肾病的免疫发病机制,泼尼松可能通过降低血清IL-17、IL-6水平参与阿霉素肾病的治疗过程。
Abstract:
Objective To explore the significance of dynamic change of serum interleukin (IL) -17,IL-6 in adriamycin(ADR)-induced nephropathy rats and the mechanism of prednisone intervention in ADR-induced nephropathy. Methods Eighty-one rats were randomly divided into control group, model group, and prednisone intervention group. ADR model was induced by a tail intravenous injection of ADR twice. The blood biochemical indexes and 24 h urinary protein, urine creatinine were measured at the end of 4, 6 and 8 weeks. The values of creatinine clearance (Ccr) were calculated by the formula. The expressions of serum IL-17 and IL-6 were detected by ELISA. Results, heavy proteinuria, hypoalbuminemia, hyperlipemia were measured in other groups at the end of 4 weeks compared with control group ,indicating that the models were successfully established. The levels of IL-17 and IL-6 in model group and prednisone intervention group were higher than control group at the end of 4 weeks(P < 0.05). At the end of 6 and 8 weeks, compared with model group, 24 h proteinuria, urine creatinine,total cholesterol, serum creatinine, blood urea nitrogen in prednisone intervention group were decreased significantly (P < 0.05), while serum total protein, albumin, endogenous creatinine clearance rate were increased significantly (P < 0.05). The expressions of serum IL-17 and IL-6 in prednisone intervention group werelower than control group(P < 0.05). Conclusion IL-17 and IL-6 may be involved in the immune pathogenesis of ADR-induced nephropathy rats. Prednisone could treat ADR-induced nephropathy by reducing the serum levels of IL-17 and IL-6.
