摘要:
目的:观察自体脂肪间充质干细胞(ADSCs)对大鼠70 %肝部分切除术(PH)后早期残肝再生的促进作用。方法:健康雄性Wistar大鼠24只随机分为两组,大鼠70 % PH组(模型组,n=12)、大鼠70% PH后自体ADSCs移植组(移植组,n=12)。比较模型组和移植组术后24、72 h血清谷氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST),获取肝组织,比较形态学改变及增殖细胞核抗原(PCNA)水平,RT-PCR检测肝细胞生长因子(HGF) mRNA表达水平。结果:与模型组比较,移植组术后24 h血清ALT水平显著降低,差异有统计学意义(t=2.925,P<0.05);与模型组比较,移植组术后24 、72 h血清AST水平显著降低,差异有统计学意义(t=4.139、3.323,均P<0.01)。光镜下观察移植组术后24、72 h肝窦轻度扩张,肝细胞结构排列紊乱程度较模型组轻。与模型组比较,移植组术后24、72 h PCNA指数显著增高,差异有统计学意义(t=3.001、2.825,均P<0.05)。与模型组比较,移植组术后24、72 h HGF mRNA表达水平显著升高,差异有统计学意义(t=2.291、3.263,均P<0.05)。结论:自体ADSCs移植可促进70 % PH后早期残余肝脏再生能力。
Abstract:
Objective:To observe the effect of autologous adipose derived mesenchymal stem cells(ADSCs) on the early liver regeneration after 70% partial hepatectomy(PH) in rats. Methods:Twenty-four healthy male Wistar rats were randomly divided into two groups:rat 70% PH(model group,n=12) and autologous ADSCs transplantation following rat 70% PH group(transplant group,n=12). Serum glutamic aminotransferase(ALT) and aspartate aminotransferase(AST) were compared between model group and transplant group at 24 and 72 h after operation. The morphological changes and proliferating cell nuclear antigen(PCNA) levels of liver tissues were compared. The mRNA expression levels of hepatocyte growth factor(HGF) were measured by RT-PCR. Results:Compared with model group,the serum ALT level in transplant group at 24 h after operation was significantly decreased,and the difference was statistically significant(t=2.925,P<0.05). Compared with the model group,the serum AST level in the transplant group was significantly decreased at 24 and 72 h after operation,and the difference was statistically significant(t=4.139,3.323,both P<0.01). Under light microscope,the hepatic sinuses in transplant group at 24 and 72 h after operation were slightly dilated,and the disorder of hepatocyte structure was significantly reduced than that in model group. Compared with the model group,PCNA index in the transplant group was significantly increased 24 and 72 h after operation,and the difference was statistically significant(t=3.001,2.825,both P<0.05). Compared with the model group,the mRNA expression level of HGF in the transplantation group was significantly increased at 24 and 72 h after operation,and the difference was statistically significant(t=2.291,3.263,both P<0.01). Conclusion:Autologous ADSCs transplantation can promote early residual liver regeneration after 70% PH.
