目的:建立并验证一种定性定量检测人全血中乙醇、甲醇、异丙醇、丙酮的方法,并将其应用于临床检测。方法:采用双流路顶空气相仪测定,色谱柱为Rtx-BAC2(30 m,0.32 mm,1.2 μm)、SH-Rtx-BAC1(30 m,0.32 mm,1.8 μm),以氮气为载气,色谱柱流量为4 mL/min,分流比为30∶1,柱温箱温度为40℃,FID温度为250℃。结果:本方法乙醇、甲醇、异丙醇、丙酮在0.062 5~8 mg·mL-1内线性关系良好,批内相对标准偏差(RSD)和相对误差(RE)分别为2.31%~10.72%和-9.50%~8.27%,批间RSD和RE分别为1.50% ~10.86%和-9.09% ~9.60%,在多种储存条件下,全血样本稳定性好。结论:本方法检测速度快、专属性强、准确度高、重复性好,本文设计了合理的标准曲线范围和质控浓度,可覆盖临床中毒检测范围。

Objective: To establish and validate a method for qualitative and quantitative detection of ethanol, methanol, isopropanol, acetone in human whole blood which can be applied to clinical test. Methods: Double flow gas chromatography was used for determination. The chromatographic column was Rtx-BAC2 (30 m,0.32 mm,1.2 μm) and SH-Rtx-BAC1(30 m,0.32 mm,1.8 μm). The carrier gas was nitrogen. The column flow was 4 mL/min, and the split ratio was 30:1. The oven temperature was 40℃, and the temperature of FID was 250℃. Results: The linear ranges of ethanol, methanol, isopropanol and acetone in this method were 0.0625-8 mg·mL-1. The intra-assay relative standard deviation (RSD) and relative error (RE) were 2.31% -10.72% and -9.50% -8.27%. The inter- assay RSD and RE were 1.50%-10.86% and -9.09%-9.60%. Whole blood samples were stable under various storage conditions. Conclusion: The method is fast, specific, accurate and repeatable. The standard linear range and quality control concentration are reasonably designed, which can cover the ranges of clinical toxic concentration.
