摘要:
目的:设计由DNA模板引导制备的纳米银簇探针,实现对肿瘤细胞中高表达的microRNA-21的定性和定量检测。方法:利用荧光光谱、紫外-可见吸收光谱、圆二色谱以及高分辨电镜表征技术,探究DNA引导合成纳米银簇的性质。通过计算血清回收率,验证探针对microRNA-21的检测效率。结果:以DNA为模板合成的纳米银簇探针能成功检测肿瘤细胞高表达的microRNA-21。结论:建立了microRNA-21的检测方法,并为临床检测肿瘤标志物提供了思路。
Abstract:
Objective: To detect the up-expressed microRNA-21 in tumor cells qualitatively and quantitatively using designed DNA-templated silver nanoclusters(DNA/AgNCs) probe. Methods: Fluorescence spectroscopy, UV-Vis absorption spectroscopy, circular dichroism spectroscopy and high-resolution electron microscopy were employed to study properties of DNA/AgNCs probes. The detection efficiency for microRNA-21 was tested by the calculating serum recovery. Results: The up-expressed microRNA-21 in tumor cells was detected by DNA/AgNCs probe successfully. Conclusion: A method for the detection of microRNA-21 is established and it could be used for clinical detection of tumor biomarkers.
